The Jak2 V617F mutation is a signaling driver for Myeloproliferative Neoplasms (MPNs), associated with the proliferation and activation of the three main myeloid lineages (erythroid, granulocytic, and megakaryoblastic). The V617F mutation site is located on exon 14 of chromosome 9, leading to an amino acid change from valine to phenylalanine at position 617. This change makes hematopoietic cells more sensitive to factors like erythropoietin and thrombopoietin. The JAK2 V617F mutation has significant diagnostic, prognostic, and therapeutic importance. Quantitative detection of this mutation is useful for monitoring patients as a residual disease marker.
