Periodic Acid Schiff (PAS) is a staining method used to identify polysaccharides such as glycogen and mucosubstances such as glycoproteins, glycolipids, and mucins in tissue. Additionally, this staining may be used to demonstrate fungal organisms in tissue samples.
Polysaccharides appear purple in staining with the PAS Stain Kit. In this type of staining, a histochemical reaction occurs in which periodic acid oxidizes adjacent diols present in sugars. As a result, the bond between two adjacent carbons is typically broken, creating a pair of aldehydes at the two free ends of each broken monosaccharide ring. These aldehydes then react with Schiff’s Solution and produce a purple color. Additionally, this kit includes a counterstain that stains cell nuclei blue.
