Description

RBC Lysis Buffer for Molecular Biology (RBC Lysis Buffer) – AP-RAD

Product Description:

The RBC Lysis Buffer for Molecular Biology by AP‑RAD is a ready‑to‑use reagent designed for the selective lysis of red blood cells (RBCs) in whole blood samples. This buffer enables efficient removal of RBCs, preserving white blood cells (WBCs) and other cellular components for downstream DNA/RNA extraction and molecular analysis. The formulation eliminates the need for toxic organic solvents or chaotropic agents, ensuring a safe and streamlined workflow.

Principle of Action:

The buffer selectively disrupts the membranes of red blood cells while keeping other blood components—such as leukocytes—intact. Many RBC lysis formulations are based on Ammonium Chloride, which induces osmotic shock leading to RBC membrane rupture. This mechanism leaves lymphocytes and other nucleated cells largely unaffected.

Laboratory Applications:

  • Preparation of whole blood samples for DNA and RNA extraction in molecular workflows

  • Improved nucleic acid extraction efficiency in samples with high hemoglobin content, which may inhibit PCR. Pretreating samples with RBC lysis enhances molecular detection sensitivity, especially when infectious agents are present at low levels.

  • Reduced sample turbidity and cleaner WBC recovery for cell biology assays or leukocyte isolation.

Features and Advantages:

  • High efficiency: Effective removal of red blood cells from whole blood samples

  • Safe formulation: No toxic solvents or chaotropic agents required

  • Wide compatibility: Suitable for human samples and molecular/microbiological workflows

  • Preservation of nucleated cells: Minimal impact on WBCs and other nucleated cells, ensuring high‑quality downstream analysis

Practical Advantages in Molecular Diagnostics:

Using RBC Lysis Buffer enhances DNA/RNA extraction yield, especially for PCR, qPCR, and other molecular assays, by minimizing the inhibitory effects of hemoglobin released from RBCs.

Conclusion:

The AP‑RAD **RBC Lysis Buffer** is an ideal reagent for blood sample preparation, nucleic acid extraction, and molecular analyses, offering a combination of safety, simplicity, and high performance for laboratory applications.
