Plasmodium are eukaryotic parasites with double-stranded DNA that cause malaria in humans. Malaria is one of the most important infectious diseases worldwide, particularly in tropical and subtropical regions, affecting millions of people annually. To date, 5 main Plasmodium species capable of infecting humans have been identified, among which Plasmodium vivax and Plasmodium falciparum are the most common causative agents of the disease. Among these, Plasmodium falciparum is the most dangerous species, accounting for the highest malaria-related mortality.

Plasmodium transmission occurs primarily through the bite of an infected female Anopheles mosquito. Additionally, transmission through infected blood transfusion, organ transplantation, and injuries from contaminated needles has also been reported. Unlike many infectious diseases, malaria cannot be transmitted directly from person to person. After entering the body, Plasmodium first infects the liver and then red blood cells, leading to the clinical symptoms of the disease.

Malaria symptoms can range from mild to severe and include periodic fever and chills, profuse sweating, headache, nausea and vomiting, muscle and joint pain, extreme fatigue, anemia, and splenomegaly. Symptom severity is greater in children, pregnant women, immunocompromised individuals, and patients with underlying diseases. In severe cases, particularly with Plasmodium falciparum infection, the disease can lead to multi-organ failure, cerebral malaria, and ultimately patient death. Therefore, rapid and accurate detection of the causative agent plays a vital role in initiating effective treatment and reducing complications and mortality.

Molecular methods such as Real-Time PCR, due to their high sensitivity and specificity, enable accurate detection of Plasmodium even in cases with low parasite load, and also have the ability for differential detection of various Plasmodium species.
