The initial laboratory diagnosis of tuberculosis and other mycobacteria depends on the direct examination of stained smears. The cell wall of mycobacteria is rich in complex lipids that prevent the penetration of common aniline dyes. However, the use of fluorochrome dyes makes these bacilli observable even under acid-alcohol conditions.

Thus, not only M. tuberculosis but also all members of the Mycobacterium genus are identified as Acid-Fast Bacilli (AFB). Fluorochrome staining is the best choice for the direct and accurate observation of these bacteria in clinical smears.
